LANCE® Ultra TR-FRET technology Basics

A simple, highly-sensitive, and highly reproducible assay platform

LANCE Ultra Assay Principle

LANCE Ultra (Lanthanide chelate excite) is a homogeneous (no wash) TR-FRET (time-resolved fluorescence resonance energy transfer) technology. One molecule is labeled with a donor fluorophore and the second is labeled with an acceptor fluorophore. Upon excitation, if they are sufficiently close for FRET (detection of the analyte by the antibodies), energy can be transferred from the donor to the acceptor fluorophore. This results in the emission of light by the acceptor fluorophore.

How LANCE technology assays work

A LANCE® Ultra assay is easy to run, and requires very few steps:

First add your analyte or sample to your microplate, then add the detection reagents: Anti-analyte conjugated Europium chelate donor and ULight™ acceptor antibody. Incubate 1-2 hours. The assay is then ready to be read on a microplate reader. When the analyte is present in the sample, the donor and ULight acceptor fluorophore are brought into proximity. Upon excitation of the donor, energy is transferred to the ULight acceptor which will emit light proportional to the level of interaction. The emission of the donor can also be detected.

LANCE TR-FRET advances your science

No wash or separation steps, extended signal stability, high sensitivity with low detection limits down to 10 pg/mL, wide dynamic range, and fast time to results. LANCE® Ultra™ TR-FRET assays are clearly options to consider when switching from WB or ELISA!
